Reactive oxygen (ROS) and nitrogen (RNS) species are produced in metabolic and physiological processes and can be overproduced in response to different stimuli. Organisms have different enzymatic and non-enzymatic antioxidative mechanisms to ameliorate the harmful effects of these oxidant species. An imbalance between oxidant species and the antioxidant defense system leads to an oxidative stress state condition that causes damage to cellular biomolecules, and it is involved in the development of several diseases. Oxidative stress can be evaluated directly by measuring ROS. Major ROS include superoxide, hydroxyl radicals and peroxides.
Since the biological effects of different oxidants are additive, measuring the Total Oxidant Status (TOS) provides a simple and reliable approach for evaluating the overall oxidative burden of a sample.
BQC Total Oxidant Status (TOS) Assay Kit is based on the oxidation of ferrous ions (Fe²⁺) to ferric ions (Fe³⁺) by oxidant molecules present in the sample under acidic conditions. The ferric ions subsequently react with xylenol orange to form a colored complex measured spectrophotometrically at 590 nm. Using a hydrogen peroxide calibration curve, the total oxidant status of the sample can be quantified.
BQC Total Oxidant Status (TOS) Assay Kit is a sensitive and reproducible assay to measure TOS in biological samples such as plasma, serum tissue homogenates or cell lysates.
