BQC NAD+/NADH Determination Assay Kit is a quick, easy, and reproducible assay for quantifying NAD+/NADH concentration in several biological samples.
The NAD⁺/NADH redox pair plays a central role in cellular metabolism and antioxidant defense:
- NAD⁺ (oxidized form) accepts electrons during metabolic reactions, becoming NADH.
- NADH (reduced form) donates electrons to the electron transport chain, driving ATP production and supporting redox balance.
The NADH present in the sample and the newly formed NADH is then oxidized by an electron intermediate acceptor resulting in the reduction of a tetrazolium salt to formazan. Formazan is a highly colored compound that shows a maximum absorption at 570 nm. The amount of formazan generated is proportional to the total amount of NAD (NAD+ + NADH) in the sample.
The native NADH can be determined alone by heating the sample (thermal degradation of NAD+) before the assay.
